[lojban-beginners] difference between di'o and tu'i

Based on the lojban dictionary I don't quite get the subtle difference between di'o and tu'i.  Can anyone expound on this for me?

di'o      BAI      at the locus of                           diklo modal, 1st place                            at the locus of ...
tu'i      BAI      associated with site                      stuzi modal, 1st place (used to situate letters)  associated with site ... ; label with location

It seems like they are similar but I don't really know what the exact definition of "locus" is anyway so maybe they're completely different.

If you wanted to say "where is John?" would you use di'o, tu'i, or something completely different?
